Developing new antimicrobials

Abstract

The alarming increase of antibiotic resistant bacteria forces us to intensively look for new antimicrobials. Our group has partially characterized a novel antimicrobial peptide, lactococcin 972, produced by Lactococcus lactis, a food-grade bacteria, which selectively kills other lactococci at nM concentrations. Moreover, no resistant strains have been found. In contrast to other bacteriocins described so far, lactococcin 972 does not target the plasma membrane. Preliminary results have shown that it could, either directly or indirectly via localized inhibition of cell wall synthesis, block septum formation and, possibly, interfere with the tightly regulated cell division event. As cell division is considered one of the prime targets for the development of novel antibiotics, Lcn972 may be considered a potential lead structure as well as a powerful tool to investigate this area in Gram positive cocci, for which very little information is available. With this in mind, at least, the following fronts should be approached: 1. Molecular structure 2. Defining its target 3. Genomic-proteomic study of its effect on sensitive microorganisms. 4. Interactions with other known antimicrobials
