Using a sterile swab or silica gel collect a specimen from the infected area.
Taking care not to contaminate the swab, return it to its tube.
Seal with adhesive tape and label using a lead pencil. Send the swab with its request form to reach the microbiology laboratory within three days.

Inoculate the swab on a plate of blood agar.
If the swab is received in silica gel ( eg. from health centre), moisten it first with sterile nutrient broth and then inoculate the plate.
Add a bacitracin disc. This will help in the identification of S. pyogenen.
Incubate the plate preferably anaerobically or in a co2 enriched atmosphere overnight at 35-370c
